Isolation of Immunoglobulins by Thiophilic Adsorption Chromatography
Izolace imunoglobulinů thiofilní adsorpční chromatografií
Izolace imunoglobulinů má význam experimentální, diagnostický i technologický. Popsaná metodavyužívá schopnosti tzv. thiofilních sorbentů značně selektivně reagovat s imunoglobuliny. Interakceje závislá na vysoké iontové síle prostředí. Po snížení dochází k uvolnění sorbovaných imunoglobulinů.Uvedeným způsobem lze jednou operací izolovat z plazmy i z dalších materiálů protilátkyv čistotě srovnatelné s jinými metodami. Výhodou metody je její šetrnost vůči imunoglobulinům,jednoduchost a láce. Postup je možno také použít pro přípravu materiálů bez obsahu imunoglobulinů.Thiofilní sorbenty jsou běžně komerčně dostupné, ale možnosti jejich využití jsou jen málo známy.

The separation of antibodies is a process of great importance. The present paper deals with themethod of thiophilic adsorption chromatography (TAC) based on the thiophilic sorbents. Thiophilicsorbents can interact with the immunoglobulins very selectively. Binding of immunoglobulins isdependent on the high ionic strength of the medium. Sorbed immunoglobulins are released by theionic strength decay. TAC provides immunoglobulins from the plasma (or other sources) in one step.Purity of immunoglobulins is comparable with results of the other methods. Benefits of TAC aredelicacy to immunoglobulins, simple handling and low costs of separation. By means of TAC it ispossible to eliminate immunoglobulins from biological materials. Thiophilic sorbents are currentlyavailable but the possibilities of their exploitation are practically unknown.
